Part 6 — The Four-Layer Synergy

How THC, CBD, d-limonene, and linalool work together as one pharmacological

strategy

Individually, each of the four compounds in the Calm & Balance formula

engages a distinct molecular pathway. But the pharmacological interaction

among them produces something that none can achieve alone — and this

has a documented mechanistic basis.

Raz et al. (2023) demonstrated in vitro that cannabis terpenes, including linalool, activated CB1 at 10–50% of THC's efficacy individually. When combined with THC at ratios reflecting natural cannabis profiles, CB1 receptor activation was several times greater than THC alone — qualifying 80 mg/day effective for GAD; antidepressant benefit confirmed; post-COVID

anxiety/depression responsive. The interaction is true pharmacological synergy, not mere additive effect.

2023 LaVigne et al. (2021) confirmed this synergy in vivo in animal models using rimonabant blockade.

THC (CB1 partial agonist): Activates CB1 in the prefrontal cortex,

amygdala, and striatum. Contributes to emotional regulation,

relaxation response, and reduced threat-processing magnitude. The

psychoactive layer, dosed for functional calm.

CBD (CB1 NAM + FAAH inhibitor + 5-HT1A PAM):

Simultaneously modulates THC's CB1 activation (the ratio is the

medicine), protects endogenous anandamide from stress-driven

depletion, and potentiates serotonergic regulation. Three

mechanisms, one compound.

D-Limonene (Adenosine A2A agonist): Activates A2A receptors

in the striatum → releases dopamine and GABA. Human RCTconfirmed

to selectively reduce THC-induced anxiety without

blocking other THC effects. The precision anxiety buffer in the

system.

Linalool (GABA-A BZD site PAM + 5-HT1A): Enhances GABA-A

inhibitory tone at the benzodiazepine site. Non-sedating at oral

doses. Six clinical trials confirm anxiolytic activity comparable to

pharmaceutical options. The calm layer that doesn't cost you

alertness.

Four pathways. Four mechanisms. One purpose: a formulation that addresses the multidimensional biology of stress and anxiety rather than targeting a single receptor and hoping for broad benefit.

This is what "formulation" means — not picking a cannabinoid because it's

trending, but designing a system because the science of how anxiety

works demands it.

Clinically Formulated Cannabinoid Therapeutics · The Formulator Is

on the Label.

Every product in the Dr. Murse Therapeutics line starts with the same question: what does the research actually say? The Calm & Balance Taffy was designed from that foundation — a 1:1 THC:CBD formulation with d-limonene and linalool, built around 20 peer-reviewed

sources and the pharmacological logic of how the human

stress system works.
